Interspecific potato somatic hybrids between Solanum malmeanum and S. tuberosum provide valuable resources for freezing-tolerance breeding

چکیده

Freezing stress affects the geographic distribution, growth, and development of potato, resulting in yield loss. Solanum malmeanum, a diploid wild species with strong freezing tolerance, was fused sensitive dihaploid S. tuberosum by somatic hybridization. In our study, 980 calli were obtained, 248 differentiated shoots obtained from calli. Parental-specific SSR markers used to analyse chromosome composition 80 randomly selected regenerated plants, obtaining 51 hybrids. Among them, 44 hybrids tested ploidy analysis years 2016 2020. During subculture, genomic levels changed due unstable 56.82% The showed better tolerance than cultivated parent. Then, freezing-tolerant backcross cultivars, we valuable breeding resources enhanced tuberization capacity similar that cultivars. correlation has no relation capacity, which indicates they are controlled independent genetic loci. transferred potato malmeanum protoplast fusion for first time, obtained.

RAPD analysis of the interspecific somatic hybrids: Solanum bulbocastanum (+) S. tuberosum.

The diploid Mexican species S. bulbocastanum (blb) was used as a source of late blight resistance in somatic hybridization with the potato (S. tuberosum, tbr) dihaploid H-8105. The produced 2x blb (+) 2x tbr H-8105 somatic hybrids did not retain the blb parent's characteristic high resistance to P. infestans. The revealed aneuploidy of blb (+) tbr H-8105 hybrids indicated a possible loss of ind...

Glycoalkaloid profile in potato haploids derived from solanum tuberosum-S. bulbocastanum somatic hybrids.

Cultivated and wild potato species synthesize a wide variety of steroidal glycoalkaloids (GA) that may affect either human health or biotic stress resistance. Therefore, GA composition must be a major criterion in the evaluation of breeding products when species genomes are merged and/or manipulated. This work reports the results of GA analysis performed on unique haploid (2n=2x=24) plants obta...

Characterization and expression of cold-induced glutathione S-transferase in freezing tolerant Solanum commersonii, sensitive S. tuberosum and their interspecific somatic hybrids.

Glutathione S-transferases (GST) form a large family of non-photosynthetic enzymes known to function in detoxification of xenobiotics. We have cloned and characterized a novel, low temperature regulated GST, Solanum commersonii glutathione S-transferase (Scgst1), from a cold acclimated wild potato species S. commersonii and studied the level of its transcription in freezing tolerant and sensiti...

Glycoalkaloid Content and Starch Structure in Solanum Species and Interspecific Somatic Potato Hybrids

When genome sections of wild Solanum species are bred into the cultivated potato (S. tuberosum L.) to obtain improved potato cultivars, the new cultivars must be evaluated for their beneficial and undesirable traits. Glycoalkaloids present in Solanum species are known for their toxic as well as for beneficial effects on mammals. On the other hand, glycoalkaloids in potato leaves provide natural...

Nuclear and cytoplasmic genome composition of Solanum bulbocastanum (+) S. tuberosum somatic hybrids.

Somatic hybrids between the wild incongruent species Solanum bulbocastanum (2n = 2x = 24) and S. tuberosum haploids (2n = 2x = 24) have been characterized for their nuclear and cytoplasmic genome composition. Cytologic observations revealed the recovery of 8 (near-)tetraploid and 3 hexaploid somatic hybrids. Multicolor genomic in situ hybridization (GISH) analysis was carried out to study the g...
